Any tutorial you want to dig into on painting in blender will tell you lots you can apply, but painting in Cycles specifically I think you just need to understand how to get the best out of painting in Blender first. I prefer to use Blender Render for the GLSL material Texture Stack, using the texture slots like layers in photoshop or gimp.
Once I do get my base painting images together, then to use them in Cycles is a matter of mixing them together properly in Color Mix nodes with the appropriate mix modes, and also setting up the stencils into the mix factor of the color mix nodes and/or the shader mix nodes. I’d point to my youtube channel, but I have been told there is too lousy of a sound problem, so you might not liek the aggravation there. I plan on updating my videos and reuploading them.
Brush creation? Depends on what you want to do - you can check out my brushes on blendswap and look at how they are made, and they aren’t specific to Blender Render or Cycles, just TexPaint Mode
