The new makehuman is long in coming (and still coming), but it will be a stand-alone application that you run just like any other.

If you have the older makeHuman 1.8 script, load the mh180b_1_1.blend file into Blender, place your mouse over the text area on the right, and press Alt-P.

It works by providing a ‘base’ mesh, a fairly androgynous character, which you manipulate using the Blender sliders on the right. Choose from the menu an area of the body to modify, such as ‘abdomen’. Then adjust the sliders to get something approaching the shape you want. Repeat ad infinitum.

Remember that you can rotate your 3D View. The only thing you can’t do is add or remove vertices.

When you’re done, save your blend file as something other than the original blend or export it. You can then load it and tweak it all you want.

Actually, the lastest beta is already stand-alone. It works much better than the plugin IMO. Just remember to strip off unused verticies after you import it into blender.
